In Vitro Screening of Bioactivities of Kjellmaniella crassifolia Extracts

  • ABSTRACT

    The biofunctional properties of hot water extract (KHWE) and ethanol extract (KEE) of Kjellmaniella crassifolia were evaluated and their potential as natural functional ingredients was assessed. Antioxidant, anticancer, antimetastatic, anti-photoaging, anti-inflammatory, and antiadipogenic activities were investigated. KEE exhibited higher 2,2-diphenyl-1-picrylhydrazyl radical-scavenging activity, nitrite-scavenging activity, and total polyphenol content than KHWE. Both KHWE and KEE inhibited the proliferation of HT-1080, HT-29, and AGS cells and suppressed matrix metalloproteinase-9 expression, suggesting antimetastatic potential. In ultraviolet A-induced human dermal fibroblasts, both extracts reduced matrix metalloproteinase-1 expression, indicating anti-photoaging effects. In lipopolysaccharide- stimulated RAW 264.7 macrophages, both extracts inhibited intracellular reactive oxygen species and nitric oxide production, as well as inducible nitric oxide synthase and cyclooxygenase-2 expression, in a concentration- dependent manner. Furthermore, both extracts suppressed lipid accumulation and downregulated adipogenic transcription factors, including peroxisome proliferator-activated receptor γ, CCAAT/enhancer-binding protein α, and sterol regulatory element-binding protein 1c. These findings suggest that K. crassifolia extracts possess diverse biofunctional properties and may serve as promising natural ingredients for functional foods and nutraceutical applications.
